Background: HERC3 (Hect domain and RLD 3) belongs to the HERC family of ubiquitin ligases which contain a HECT domain and at least 1 RCC1-like domain (RLD). E3 ubiquitin-protein ligases accept ubiquitin from an E2 ubiquitin-conjugating enzyme in the form of a thioester and then directly transfer the ubiquitin to targeted substrates. The RLD domain is predicted to act as a guanine nucleotide exchange factor for small G proteins.
