Background: c-Kit (CD117, Steel factor receptor, and stem cell factor receptor) is a 145 kDa glycosylated transmembrane protein whose extracellular domain contains Ig-like domains that bind c-Kit ligand (KL; stem cell factor). The interaction of c-Kit and its ligand is required for normal hematopoiesis, mast cell differentiation, melanogenesis, and gametogenesis. Mutations in KIT are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism. Phosphorylation at tyrosine 721 of c-Kit allows binding and activation of PI3 kinase.
