Background: The STAT 4 protein is a transcription factor activated by hormone and cytokines. STAT 4 is most similar to STAT 1 (52%) and STAT 3 (47%). Functionally STAT 4 is similar to other STAT family members in that it can be tyrosine phosphorylated by JAK 1 or JAK 2. STAT 4 forms homodimers and heterodimers with related STAT family members. Tyrosine phosphorylated STAT 4 can bind the interferon g activated site (GAS), and serine phosphorylation is also required for maximal transcriptional activity. STAT 4 expression is restricted to the thymus, spleen, and testis.
